The way you place or keep your guitar doesn't really effect on the strings.. However sometimes the strings might be out of tune accordingly with weather.. But if you don't touch the tuning pegs doesn't effect the strings tuning. ( in case of electric guitar, when you used some bending it might run out off tune and when using floating bridge tremolo might effect the strings too).

It’s been done but it doesn’t work very well. Steel string acoustic and electrics are intonated for right hand stringing. That means the scale length for each string is a little bit different. If you string it left handed the heavy strings will ring progressively sharper as you go up the neck and the light strings will ring flatter as you go up the neck. It’s generally not a problem with classical guitars but now days even some of them have intonation built into the bridge saddle, Jasmine nylon string guitars for example. Another problem is that some acoustic steel string guitars are braced a little heavier on the bass side and lighter on the other. Those guitars may sustain damage on the lightly braced side if you string it left handed. On electrics the scale length adjustment in the saddles can only partially make up for this so they still will not tune up right.
